Imaging Methods

About us

Introduction

IMCF covers three main areas-optical (fluorescence) microscopy, electron microscopy and flow cytometry. We offer our users training on and access to high end instrumentation, plus we provide consulting services on optimal experimental design, sample preparation, data acquisition and analysis and interpretation of the obtained results.

Acknowledgments

All users are required to acknowledge IMCF in any publication that uses data acquired in the facility. The more acknowledgments our facility gets, the more we will be able to donate the running costs (lower instrument fees), maintain and upgrade the equipment, and thus offer you better services for your own research. Thank you very much in advance!

As 27% of our running cost for optical and electron microscopy is currently covered from Czech-BioImaging large infrastructure project LM2015062, to get the discounted fee please acknowledge this project as well. Thank you!

    Intravital inverted two-photon and confocal microscope with 34 channel spectral detection and with four reflected and two transmitted NDD detectors (including two highly sensitive GaAsP detectors suitable for FLIM), equipped with MP laser with OPO and with the full set of visible cw lasers.

  • FEI Helios NanoLab G3 UC–FIB-SEM

    Scanning electron microscope with focused ion beam milling equipped for cryo-imaging and correlative light and electron microscopy (CLEM) plus accessories (Leica EM HPM100, ULTRACUT UC7 with FC7, AFS2, CPD300 A and KMR3) for sample preparation.

  • BD FACS Aria Fusion

    (5 lasers, 19 detectors, jet-in-air 4-way sorter, 70 000 cells per minute) and BD LSR Fortessa SORP (5 non-collinear lasers, 20 detectors, 40 000 samples per second) for complex fluorescence flow cytometry analysis and sorting.

  • Super-resolution Nikon Ti-E microscopes with N-SIM and N-STORM modules and Nikon Ti-E with STED module from Abberior Instruments

    Each module allows for two-colour 3D super-resolution acquisition, using different approaches to break the diffraction limit of optical resolution.

  • Leica TCS SP8 DM6 CFS and Leica DMi8

    Upright confocal and inverted wide-field microscopes for combined optical and electro-physiological measurements, Nikon Ti-E with Laser H-TIRF System for long term live cell observations and Carl Zeiss AxioZoom.V16 for large specimen observation.

  • Leica TCS SP8 WLL SMD-FLIM

    Inverted confocal microscope with a fluorescence lifetime imaging module (FLIM), including pulsed white light laser (470 nm – 670 nm), pulsed 405 nm laser and freely spectrally selectable detection with two high sensitivity HyD detectors.

CF SERVICES

In fluorescence microscopy, we cover the following methods: wide-field and confocal microscopy, two-photon deep intravital microscopy, 3D two-colour super-resolution microscopy (STED, PALM, STORM, SIM), functional imaging (spectral, FLIM, FCS), TIRF, live cell imaging and combined electrophysiological and optical acquisition. In electron microscopy, we offer 3D SEM-FIB acquisition on embedded, dried or frozen samples, elemental analysis (EDS) and combined light and electron microscopy (CLEM) approaches. In flow cytometry, we offer fluorescence activated cell sorting (FACS) and analysis. The strength of our facility is the application, implementation and development of advanced fluorescence imaging techniques, especially super-resolution and functional (FLIM, FCS) imaging.

POTENTIAL FOR COOPERATION

We are seeking for key partners with biological,  medical and also material science projects that could benefit from custom optimized advanced imaging techniques. We also welcome industrial partners not only interested in using our high-tech equipment, but also partners needing a test facility for their new products and partners interested in commercialisation of our newly developed methods from the field of super-resolution and functional imaging.
